A starting rituximab administration establishes circulating exposure. Exposure declines through nonspecific and B-cell-dependent loss and drives depletion of the circulating CD19-positive B-cell pool. Baseline production and natural turnover govern B-cell recovery. Tissue pools, explicit CD20 binding, repeat dosing, and clinical benefit are outside the model.
